Research Description: Diseases or disorders affecting the anterior and posterior segments of the eye, such as keratitis, diabetic retinopathy, age related macular degeneration and retinitis pigmentosa, are the leading causes of decreased visual acuity and blindness. Successful treatment modalities require delivery and maintenance of therapeutically effective drug concentrations at the target sites. Drug delivery to the ocular tissues is, however, limited by several physiological processes. Topical instillation is challenged by tear flow, structural complexity of the cornea, long diffusional distance, counter-directional intraocular convection and conjunctival, scleral and choroidal vascular and lymphatic systems. Although systemic administration offers an alternative route, the blood-ocular barriers restrict the diffusion of several therapeutic agents (especially hydrophilic compounds and efflux pump substrates) from the systemic circulation into the ocular chambers. My primary area of research focuses on the development of noninvasive or minimally invasive drug delivery strategies to enhance ocular bioavailability of poorly permeating compounds through
- Biopharmaceutical and pharmacokinetic considerations
- Prodrug design for the modulation of physicochemical characteristics or the targeting of nutrient transporters
- Modulation of efflux mediated by P-glycoprotein, MRP and other efflux proteins
- Evaluating effect of drug-drug interaction on ocular pharmacokinetics, metabolism and efflux
- Developing novel ocular drug delivery platforms
